Abstract
Experiments in curarized rabbits with implanted electrodes: all drugs were given i.v. 1) LSD (20 mcg/kg) produces and EEG pattern of activation with desynchronization of the cortex and hypersynchronization on the hippocampus. Mescaline and bufotenine have an effect similar to that of LSD. The effect of all 3 drugs lasts for a long time. 2) Harmine (0.4 mg/kg) and nalorphine (2.0 mg/kg) desynchronize the cortex also but alternately with synchronization. 3) WIN-2299 produces a sleep EEG which is also not interrupted by painful stimuli. A similar effect is observed with the drugs No. 329 and No. 336. 4) Phencyclidine: 1 mg/kg produces on the motor cortex long wave delta-activity alternating with rapid beta waves, 2 mg/kg in all leads slow synchronized activity which changes over to the alert type EEG after painful stimulus. . Site of action; relations to the psychic effect in man: LSD, Mescaline, bufotenine, harmine and nalorphine stimulate the formatio reticularis (FR). The changing EEG of rabbits after harmine and nalorphine corresponds in man to the change of alerting and suppression. While LSD and Mescaline do not influence the consciousness, the state after harmine or nalorphine stimulates an organic syndrome with disturbances of consciousness. The anticholinergic WIN-2299, drug No. 336 and No. 329 inhibit the FR and produce also an "organic" state in men. Phencyclidine affects the cortex, it does not cause hallucinations and illusions, but peculiar disorders of thinking. . Comment: WIN-2299 = (2-diethylaminoethyl cyclopentyl-(2-thienyl)-glycolate hydrochloride). Drugs Nos. 336 and 329 are esters of piperidine and benzilic acid. Nalorphine = Nathin. Phencyclidine = Seryl.
