Abstract
The effect of LSD on immunoglobulin biosynthesis was studied using anti-fluorescyl antibody producing lymphoid cells isolated from rabbits. Method Antifluorescyl antibody producing lymphoid cells (spleen and lymph node) isolated from adult albino rabbits, were incubated in minimum essential medium (MEM) containing 3H - leucine (medium I), 3H-leucine plus 14C-tryptophan (medium II) or as medium II plus USE) (0.1-10.0 fig) (medium III). Following dialysis the extracellular supernatants were analysed for radioactive label incorporation into antibody protein. Results Total amount of 3H-leucine incorporation ... media I and II was almost identical, indicating that when the tryptophan content of MEM was replaced by '4C-tryptophan, protein synthesis was not altered. LSD depressed 14C-tryptophan incorporation into secreted protein as evidenced by increased 3H-leucine to 14C-tryptophan ratios. Sucrose gradient analyses, isoelectric focussing, etc. indicated that the extracellular labelled material of media I and II was 80-90 0mmunoglobulin. Extracellular material from medium III was of lower molecular weight and had little or no anti-fluorescyl antibody; sucrose gradient analysis revealed that no 7S antibody was being secreted from the cells incubated in presence of LSD. Conclusion LSD interferes with tryptophan incorporation into antibody protein.
