Abstract
The effects of various psychotomimetic drugs and cold water swimming stress on the metabolism of 3H-norepinephrine (3H-NE) in rat brain were investigated. Method Male Sprague-Dawley rats (160-250 g) were lightly anesthetized with ether and injected, 1, intracisternally 25 111 with 8.3 micro-c H-NE (6.88-7.76 c/mmol) and 90 min later subjected to cold water swim stress or i.p. drug or saline (0.9%) injections or 2, i.v. with L-tyrosine-3,5-3H (80 pc) (20 c/mmol) in 0.9aline. Drug or control - (saline) injections were given immediately; cold water stress was imposed 40 min later. 1300 mcg/kg LSD Reduced levels of 3H-NE and endogenous norepinephrine (NE) but the latter had returned to control levels4 hr after LSD. Metabolites of 3H-NE were at or above control levels but there was no differentiation in the pattern of 3H-NE. A dose response curve for LSD (260-1300 mcg/kg) revealed that only 1300 mcg/kg caused consistent alterations in catecholamine degra-dation. Psilocybin 25 mg/kg reduced 3H-NE and endogenous NE content dramatically. 3H-Normetanephrine (3H-NMN) levels were increased considerably. Other metabolite levels were increased slightly. After 25 mg/kg mescaline 3H-deaminated catechol metabolites were elevated 20 min after injection but declined rapidly in contrast to 3H-NMN levels which increased slowly. Cold water swim stress caused increase in all metabolites except 3H-NMN. Endogenous NE was decreased. 2 mg/kg i.p. D-Amphetamine sulphate injected 2 hr after 3H-NE increased 3H-NMN but did not alter other metabolites. Only LSD altered dopamine levels and this failed to increase dopamine specific activity
