“An Analysis of Some Discriminative Properties of d-Amphetamine”.

Abstract
Some discriminative properties of dexamphetamine (A, K+K) are reported. Rats (280-350 g) were trained to discriminate between the drugged :state (A) and the-non - drugged state (saline) on a 2-lever response schedule. A dose -response study of the discrimination to gradedA doses (0.0625-2 mg/kg) was performed. The effect of time delaying on the A discrimination cue was determined in rats given 1.0 mg/kg and tested 60, 90, 120 or 150 min later. Rats trained for the A cue were given psilocybin 0.5 or 1.0 mg/kg, THC 0.5 or 1.0 mg/kg, mescaline (Sigma-Chem.) 5.0 or 10.0 mg/kg, LSD (LSD-25) 0.04 or 0.08 mg/kg, methamphetamine (Sigma-Chem.) 1.0 mg/kg or caffeine (Sigma-Chem.) 6 and 20 ma/ kg and the discrimination behavior was assessed in relation due saline and A performances. Also the effects of a-methy -p-tyrosine (a-MT, Merck-USA) 200 mg/kg p.o. pretreatment on the A-saline discrimination after A 0.50 mg/kg were found. A-saline discrimination reached 180% after 13 days training. Responses to A 0.0625-2 mg/kg were mainly on the saline lever at doses 0.25 mg/kg. Above this dose, the response indicating recognition of a drugged state increased dose-relatedly to 81% with A2.0mg'/kg. When trials were delayed for-60-min post-dose, a 74% response on the A lever was obtained. A 90 min delay decreased A lever response to­50%. After 120 and 150 min responses on the A lever were 33% and 1-4% respectively. After 30 min latency a 94% A lever response was reattained. Of all the drugs tested only methamphetamine caused a discrim-nation response comparable to that obtained with A. All other drugs induced a preferential saline fever response. a-MT Reduced the A:lever- response to 40% (control 760) after A 0.5 mg/kg.
