“Hyperthermia- and L-Tryptophan-Induced Increases in Serotonin Turnover in Rat Brain”.

Abstract
Several lines of evidence indicate that hyperthermia increases 5-HT turnover in rat brain: (1) Corrodi, Fuxe, and Hökfelt (1967) found that the rate of 5-HT depletion from rat brain was increased by exposure to high environmental temperature after inhibition of tryptophan hydroxylase by o-propyl dopacetamide. (2) Aghajanian fund Weiss (1968), as well as Reid, Volicer, Smookler, Beaven, and Brodie (1968) showed That exposure.to high environmental tempera - 'ture increased the concentration of 5-HIAA in brain without increasing that of 5-HT. (3.) Weiss and Aghajanian (1971) found that hyperthermia increased the rate of firing of 5 - HT-containing raphe neurons. Tagliamonte, Tagliamonte, Perez-Cruet, Stern, and Gessa (1971) confirmed the increase in rat brain 5-HIAA during hyperthermia, but found that this condition also increases the concentration of L - tryptophan. Since it is well established that l-tryptophan loading greatly increases the concentration of 5-HIAA in rat brain (e.g., , Eccleston, Ashcroft, and Crawford, 1965) the possibility arises that the increase in 5 - HIAA after hyperthermia might be, at least in part, a secondary effect mediated by an increase in L-tryptophan concentration Furthermore, Aghajanian (1972) has recently reported that l -tryptophan loading inhibits the activity of raphe neurons. The present experiments were carried out to evaluate whether increased cerebral L - tryptophan concentration and increased serotonergic nerve activity are responsible for the elevation of 5-HIAA concentration' in rat brain, during hyperthermia.
