“In vivo changes of guanosine 3',5'-cyclic phosphate in rat cerebellum by dopaminergic mechanisms”.

Abstract
The effects of dopaminergic mechanisms on guanosine 3',5'cyclic phosphate (cGMP) cerebellar concentrations were studied in rats. A dose-dependent increase in cerebellar cGMP occurred in rats (160-180 g) 30 min after apomorphine treatment (1.0-10 mg/kg, i.p.). Peak cGMP levels occurred at 15 min after dministration and had returned to control levels after 2 hr. Haloperidol (2.0 mg/kg), itself inducing a decrease in cGMP abolished the apomorphine response if given 30 min before this drug. Reserpine (3,10 mg/kg, s.c.) also reduced cGMP levels in the cerebellum (by 25 and 50%) 16 hr after its administration, and decreased the apomorphine induced increase by 40%. Hyoscine (iO mg/kg, i.p.), itself without effect on cGMP level, abolished the apomorphine response. Methylatropine was without effect on the apomorphine activity, while LSD (0.5-2.0 mg/kg) produced a dose-dependent increase in cGMP comparable to that of apomorphine. Its action was abolished by pretreatment with haloperidol (2 mg/kg). However, 2-Br-D-lysergic acid diethylamide had no effect on cGMP concentrations in the cerebellum. Dopamine receptor stimulation and blockade elevate and reduce the content of cGMP in the cerebellum.
