Abstract
The binding of 5-hydroxytryptamine (5HT) and LSD to bovin cerebral membranes in vitro was studied. Methods Fresh bovine cerebral membranes from various sites were homogenized and centrifuged; the coarse mitochondrial fraction obtained was fractionated variously according to previously des cribed methods. The various fractions were then incubated with - 3H-5HT and/or 3H-LSD and filtered; the radioactivity remaining in the filter was measured. Results 5HT was bound to crude brain membrane preparations with different affinities (KD = 1 to 2 x 10 q M for the highest and KD 1 to 2 x 10 3 M for the lowest). LSD also had 2 binding affinities with a similar degree of difference between them (KD = 3 to 4 x 10-9 M and KD = 2 to 3 x 10 M). In microsomal membranes both affinities were represented but in purified synaptosomal membranes only the higher affinity binding sites were present. The high affinity binding sites for 5HT and LSD were different, since the 2 did not compete for them. On microsomal membranes, there was a direct relationship between the low affinity binding for 5HT and high affinity binding for LSD
