“Depressor Affect of Kynurenine and Its Metabolites in Rats”.

Abstract
The effects of kynurenine and its metabolites on blood pressure in rats were studied. Methods Rats (160-240 g) were anesthetized with urethane (1.75 g/kg) and blood pressure measured via the carotid artery. Kynurenine and its metabolites were given i.v. Results The threshold dose of kynurenine which produced a definite reduction in blood pressure was 0.2 ng; threshold values for other test drugs were picolinic and quinolinic acids (1 mg), 3hydroxy-kynurenine, 3-hydroxy-anthranilic acid, anthranilic acid and nicotinic acid (2 mg). The duration of the hypotensive effect of kynurenine (0.2 ng) was 5-7 sec but this increased with dosage; a peak reduction in BP of 20-32 mmHg was given by doses of 100 lug. Doses of 400-2000 )lg kynurenine gave a 10-15 see duration reduction (4-6 mmHg) in blood pressure followed by a rise of 12-16 mung which lasted for 2-3 min. The response to kynurenine (20-200 ng) was similar tothat produced by acetylcholine (25 ng) and serotonin (5 ng). The hypotensive effect of kynurenine was unaffected by bilateral vagotomy, atropine (10 mg/kg i.p.) methysergide (50, 100 mcg) LSD-25 (10-25 mcg) or 1,6-dimethyl-8p-carbo-benzylkydroxyaminOmethyl- 1 O -alpha- e rgolin. At doses of 5-7 mg, kynurenine metabolites showed the following order of decreasing effectiveness; nicotinic acid, quinolinic acid, 3-hydroxy-kynurenine, anthranilic acid, picolinic acid and 3-hydroxy-anthranilic acid.
