Abstract
Approaches to the analysis of drug interactions at the genetic level are reviewed. Most studies made have focused on the psychomimetic drugs such as marijuana and LSD. Molecular interactions with DNA, chromatin or mitotic spindle components are a useful way of assessing the potential for genetic damage and studies of this type involving LSD, tetrahydrocannabinol (THC) and harmine are reviewed. Effects may be manifest as alterations in DNA replication., repair of transcription. Workwith marijuanacigarettes, LSD, UV-irradiation and THC is described to illustate these factors. Testing for mutagenic effects may involve S. typhimurium histidine auxotrophs in vitro or an in vivo modification of the assay which has the advantage that it permits the effectsof metabolites of the test drug to be evaluated in the same system. Tests done on this system have detected a mutagenic potential for cigarette smoke condensates and benz(a)pyrene. Induction of aryl hydrocarbon hydroxylase, an enzyme that may produce carcinOgenic epaxide derivative of polycyclic hydrocarbons, has been shown to occur with THC and cannabis. Other studies with LSD and THC are also reviewed.
