Abstract
The interaction of delta-9-tetrahydrocannabinol (THC) with reserpine, phenobarbital and LSD-25 on plasma and adrenal corticosterone was studied. In acute studies groups of male albino rats (120-150 g) were given THC 10 mg/kg i.p. followed immediately by phenobarbital 75 mg/kg (May+Baker), reserpine 2.5 mg/kg (CIBA-Geigy) or LSD-25 50 mcg/kg (Sandoz) or alternatively THC 50 mg/kg or any of the other drugs alone. In subchronic studies animals received THC 10 mg/kg/day for 21 days; a number of the animals were also given reserpine, phenobarbital or LSD-25 with the last injection of THC on day 21. All the rats were sacrificed by decapitation at intervals up to 360 min after the last injection. Trunk blood was collected and the adrenals were removed. THC alone produced a dose dependent stimulation of ACTH release, the peak response occurring 45-90 min after adminitration. Phenobarbital lowered the basal corticosterone concentrations and partially inhibited the corticosterone elevating effect of THC. Administration of LSD-25 did not alter the levels of corticosterone. Reserpine raised plasma and adrenal corticosterone concentrations; when administered with THC an additive effect was observed, which may be due either to additional release of ACTH or to increased corticosteroid output. Chronic administration of THC did not produce any tolerance either to the effects of THC alone or to single doses of the other drugs.
