Abstract
The effect of repeated injections of LSD on rat brain 5-hydroxytryptamine (5HT) and 5-hydroxyindoleacetic acid (5HIAA) levels was studied. Methods Male Sprague-Dawley rats (85-100 g) were injected with LSD (100 mcg/kg, i.p.) once daily for 4 wk. or LSD daily for 2 wk and then saline for 2 wk. At the end of treatment, 5HT and 5HIAA were measured in the cerebellum, cortex, midbrain, ponsmedulla, striatum and remainder of the brain. Treatment with LSD for 4 wk caused an increased turnover of 5HT in the cerebellum and an increased 5HT level in the midbrain. The shorter treatment schedule caused an increase in 5HT in the cortex and midbrain and no change in 5HIAA levels in any region. Conclusions Treatment with LSD for 14 days caused an increase in 5HT in certain areas of the brain without a change in 5HIAA. If treatment was continued for 4 wks, smaller effects were observed, indicating that tolerance may have occurred.
