Abstract
The effects of injection of ergometrine (E) and other ergot derivatives into the nucleus accumbens on locomotor activity were studied. Methods Rats (200 + 20 g) were implanted with cannulae into the nucleus accumbens. Intracerebral injections of the drugs were given bilaterally. Locomotor activity was recorded continuously before and for at least 5 hr after the intracerebral injection. Results Bilateral injection of E (1 mcg) into the nucleus accumbens markedly stimulated locomotor activity. Propranolol, phenoxybenzamine, and methysergide (all 5 mg/kg i.p.) had no clear influence on the effect of E. Peripheral injection of E (2 mg/kg i,p.) immediately after intracerebral injection of E resulted in the sudden death of 8/9 rats. Scopolamine (1 and 15 mg/kg i.p.) produced and initial enhancement of the effect of E on locomotor activity, Pretreatment with reserpine (Serpasil, 2 mg/kg i,p.) alone or in combination with a-methyl-p-tyrosine methylester (100 mg/kg i.p.) potentiated the effect of E and reduced the time of latency of the E effect. Locomotor stimulation was reduced when L-dopa (50 mg/kg i.p.) and Ro-4-4602 (50 mg/kg i.p.) were administered prior to E, but not after pretreatment with 5-hydroxytryptophan (50 mg/kg i.p.) and Ro-4-4602. Apomorphine (2 mg/kg i.p.) administered about 10 min after onset of E-induced locomotor stimulation produced within 5-10 min stereotyped behavior lasting about 1 hr. Bilateral injection into the nucleus accumbens of haloperidol (Serenase, 2.5 ug) or (3,4-dibydroxyphenylamino)-2-imidazoline (DPI) (5mcg markedly inhibited the effect of E. Clonidine (5 1lg bilaterally) reduced the effect of E only slightly. Injection into the nucleus accumbens of DPI alone or of ergocornine, bromocyrptine (both Sandoz, 5 and 10 mcg), LSD dibydroergotamine (Sandoz), and methysergide (all 2 and 5 mcg) did not produce a clear locomotor stimulation.
