Abstract
The effect of LSD on raphe unit activity was studied. Male Sprague-Dawley rats were pretreated with LSD tartrate 100 Ámcg/kg or 1000 mcg/kg per day 4 days, 1000 mcg/kg per day for 7 days or 250 mcg/kg 6 holy for 4 days, or with saline. Recordings were made of the activity of dorsal and median raphe nuclei between 16 and 24 hr after the final injection of LSD or saline. For the recordings, glass micropipettes filled with 2 M NaCl saturated with fast green (impendances ranging from 4 to 6 my, measured at 1 kHz) were stereotaxically lowered into the raphe nuclei, under chloral hydrate anesthesia (400 mg/ kg) There were no significant changes in the dose-response relationships between raphe-unit activity and LSD administration following any of the dose regimens. Conclusion The dramatic tolerance which developed to LSD on behavioral tests is not mediated by a change in the sensitivity of serotonin containing neurons to LSD.
