Abstract
Quipazine (QU) discriminative stimulus properties in rats were studied. Methods QU (2.5 mg/kg) or saline was injected into Sprague-Dawley rats (275-320 g). 30 Min later they were conditioned to receive H2O by pressing one of 2 bars. Different bars were used after drug or saline injection. When discrimination was above 85%, the dose QU (0.31-1.25 mg/kg) before testing or the delay before testing (5-240 min) was varied. Some trained rats received damphetamine (1 or 2 mg/kg), apomorphine (0.5 or 1 mg/kg) or D-LSD (0.02-0.10 mg/kg) instead of QU while others received methysergide (1 mg/kg), cyproheptadine (1 mg/kg), methiothepin (1 mg/kg), fluphenazine (0.5 or 1 mg/kg) or haloperidol (50 or 100 mcg/kg) 30 or 60 min before QU. Results Discrimination by QU-treated rats reached 80% after 12 training sessions, Discrimination fell after doses of QU below 0.94 mg/kg. The onset and duration of action of QU were determined to be about 5-15 min and 60-90 min. respectively. The QU cue could only be transferred to LSD (0.08 and 0.10 mg/kg) and discrimination was blocked by methysergide (71%) cyproheptadine (28%) and methiothepin (25%). LSD and QU have a common mechanism of action which may (or may not) involve serotonin; a role for dopamine is not indicated.
