Abstract
The study examined the effects of methergoline (MTG) on the behavioral disrupt“on produced by hallucinogens: d-Lysergic acid diethylamide (LSD; 12.5-400 mcg/kg) and N,N-dimethyltryptamine (DMT; 1-8 mg/kg) as examples of indolealkylamines, and 2,5-dimethyl-4-methylamphetamine (DOM; 0.125-4 mg/kg) and mescaline (5-28.4 mg/kg) as phenethylamines. Food deprived male rats were taught to press a bar on a fixed ratio-40 (FR-40) operant schedule for food reinforcement. The hallucinogens given i.p. resulted in a dose-dependent cessation of responding for some period of the 40 min test session ('hallucinatory pause'), MTG (1 mg/kg, 180 min before the FR-40 session) completely antagonized pause-producing effects of DOM and mescaline, even with supramaximal doses. The block of LSD and DMT effects was reversed when the doses of these agents were increased sufficiently. These data support the hypothes“s that 5HT neuronal systems are important in the behavioral effects of hallucinogens. There appears to be subtle differences in the mechanisms by which the phenethylamine and indolealkylamine hallucinogens produce their behavioral effects.
