Abstract
The effects of mescaline and LSD on lymphocyte function was studied in-vitro. Mescaline was incorporated into protein, during incubation with immune lymphoid cells in vitro. Protein synthesis inhibitors did not prevent this incorporation and no effect on antibody synthesis was found. Mescaline inhibited incorporation of thymidine into DNA in mitogen stimulated lymphocytes when present at concentrations between 10-200 mcg/ml. Nucleoside transport was also inhibited at high mescaline concentrations (400 mcg/ml) but transport inhibition was insufficient to explain the inhibition of thymidine incorporation into DNA. Competitive binding experiments with beef brain tubulin, radioactive colchicine and mescaline indicated mescaline and colchicine bound to tubulin at the same site. Radioactive mescaline bound to tubulin with a dissociation constant of about 5 x 10^-4 M. In vitro polymerization of tubulin was measured by light scatter and by a filtration assay. Using partially purified microtubule preparations, mescaline inhibited polymerization at low protein concentrations and caused enhanced polymerization at high protein concentrations. Mescaline caused a transitory reversal of colchicine inhibition of tubulin polymerization Anti-tubulin antibodies showed mescaline was associated with tubulin during in vitro cell incubations. Polyacrylamide gel electrophoresis in the presence of SDS showed mescaline was associated with proteins larger than 50000 Daltons during in vitro cell incubations. LSD had no effect on thymidine incorporation into mitogen stimulated lymphocyte cultures or on microtubule polymerization. Mescaline effects were not comparable to earlier effects found for LSD.
